点击化学("click" chemistry)是诺贝尔化学奖得主Sharpless教授在2001年提出的一种快速共价偶联策略。 而"光点击"与"点击"化学的关系是青出于蓝,胜于蓝。它继承了点击反应的优点,同时也是受光激发,实现时空控制构建共价连接的化学利器。 在“光点击”化学反应中,利用直接光转化或者光催化策略,实现生物正交偶联的精准光控,达到亚细胞/单分子级别的时空精准度。采用环张力环融合策略,将“光点击”反应提高10倍以上的反应动力学。利用课题组原创的环偶氮亲偶极体,首次合成DBTD丙氨酸作为非天然氨基酸衍生物,并将其用于遗传密码扩展(GCE)编载重组蛋白质表达,实现超快速活体内蛋白质特定残基的荧光标记。这一系列创新研究,将推进“光点击”生物正交反应在活体动物中的靶标分子的跟踪和操控研究,起到了中流砥柱承前启后的作用。

主要工作业绩:
余志鹏博士毕业于四川大学,师从冯小明院士。后赴美国纽约州立大学布法罗分校化学系开展博士后研究工作,后升任美国纽约州立大学布法罗分校化学系研究助理教授。在入职四川大学以来,致力于研究以“光点击化学”为基础的生物正交化学以及其在活体生命环境中的外源性化学功能的引入及其相关的应用,致力于开拓化学生物学前沿的研究方向的教学和科研工作。
主要开展前沿的“光点击”化学反应的原创性开发,以及其在可光控的分子生物学探针研发,原生活体中生物分子的生物正交标记、示踪(基于超高效、超高选择性的有机化学反应为基础)中的突破性创新。研究高度生物兼容且精准可控的超快共价偶联(脱保护)反应、实现超高化学专一性的生物正交偶联反应策略。余志鹏课题组已经研发出国际前沿的两代、三类型的“光点击”反应,其可在高度拥挤的活体细胞浆质内实现选择性的生物大分子修饰和功能调控,对生物分子添加外源性功能性残基,扩展活体生命系统的功能。该研究不仅仅可以通过化学功能团的载入实现对特定生物代谢,基因的调控,细胞的信号传导等等生物过程的干预和操控,同时也是精准药物发现和原位构建3D生物功能材料的基础化学利器。
建立了以介离子“悉尼酮”为基础的“光点击”化学新方法,丰富了“光点击”化学前沿工具库,达成可控、高效构建共价键,以及协同断键释放功能。实现其在活体中原位精确标记生物分子以及药物分子释放的诊疗一体化交叉应用。原创发现一类T-型“光开关”环偶氮分子,其作为独特的亲偶极体与悉尼酮配套组建新一类“光点击”反应系统,验证了这一全新生物正交报告基的全能性,提出从光能利用的角度来突破光点击化学发展的概念。定向进化MmPylRS(吡咯赖氨酸tRNA合成酶),创新发展环偶氮非天然氨基酸的遗传密码扩展系统(GCE),实现活细胞中蛋白高精度示踪以及光控基因表达的新思路。
