The works in my group mainly focus on developing different reaction-based probes or nanomaterial probes and their application in cell imaging. We have developed a series of subcellular-organelle-targeted probes for imaging and detection of reactive oxygen species (ROS), reactive nitrogen species (RNS) and reactive sulfur species (RSS) in living cells. Meanwhile, we have designed purine-based AIEgens which exhibit lipid droplets specific property in live cellular imaging; based on this skeleton, a wash-free imaging and ultrafast staining process for plasma membrane was developed with good biocompatibility. Currently, we are developing near-infrared (NIR) fluorescent dyes with large stokes shift, including phosphorus-substituted rhodamine and sulfoxide-substituted rhodamine, and investing their application in autophagy and fluorescence-guided surgery. We also focus on selective distinguish and detection of modified bases in DNA methylation. By exploring reaction-based labelling/fluorescent reagents, we have realized selective detection of 5fC and 5fU, respectively. We are developing a photodissociation technique for selective enrichment of modified bases and single base resolution sequencing.
