1981-1988   兰州大学化学系有机化学专业学习,分获理学学士学位和理学硕士学位。

1993-1996   中国科学院化学研究所学习,获理学博士学位。

1988-1993   西南师范大学化学系从事有机化学教学和科研工作,历任助教和副教授。

1996-2000   中国科学院成都有机化学研究所从事不对称催化反应研究,历任副研究员和研究员,不对称合成联合开放实验室副主任,于1999被评聘为博士生导师。

主要工作业绩
手性是自然界的基本属性。手性化合物在医药、农药、食品添加剂、信息素以及功能材料等方面起着重要作用,其社会需求日益增大。不对称催化是获得光学活性化合物的最高效方法,具有原子经济性和广阔的应用前景;其核心科学问题之一是发现和发展新型高效、具有广泛通用性的催化剂和不对称反应。

      手性催化剂是不对称催化的关键,通常认为配体或者催化剂的骨架具有刚性是优势手性催化剂的重要条件,许多配体或催化剂一般只适用于个别反应和部分底物,不具通用性。我们设计合成了具柔性直链烷基链接的C2对称双氮氧酰胺化合物,建立了结构多样性、可满足不同反应需求的手性双氮氧-金属络合物催化剂库,是目前对反应类型和底物最具广谱性、价格最低的催化剂之一。这是一类具有自主知识产权的新型优势手性配体,突破了对配体刚性骨架的传统要求,获得了具有最佳手性口袋效应的手性催化剂,为新手性催化剂的设计提供了新思路。此外,运用双功能催化、分子识别、自组装、不对称活化等新策略和新概念,建立了胍-酰胺和二胺类双功能有机催化剂库和组合配体金属络合物催化剂库。利用这些自己发展的上述催化剂,高效高选择性地实现了30多类重要不对称催化反应。该类催化剂还显示了一些独特的催化性能,实现了7类具有挑战性的不对称催化新反应,其中一类反应被Elsevier出版社的有机人名反应专著冠名为“Roskamp–Feng”反应。
现已分别在Acc. Chem. Res. (1), Chem. Rev. (1), J. Am. Chem. Soc.(10), Angew. Chem. Int. Ed. (19), Chem. Eur. J. J. Org. Chem., Org. Lett.Adv. Synth. Catal. Tetrahedron, Synlett等刊物上发表论文200多篇,SCI他引超过3800次。其中J. Org. Chem., 2002, 67, 2275这篇论文,被美国化学会的Chemical Reseach NewsHeart Cut收录,并对此作了专门的正面评论;J. Am. Chem. Soc., 2005, 127, 12224这篇论文入选了该刊200579月的Most Accessed Articles(20篇论文); J. Org. Chem. 2007, 72, 7715被评为该刊物的200779月的Most Accessed Article; J. Org. I hope that this catalyst system can be applied to many other asymmetric syntheses”;同时自然杂志的新闻刊物Nature China对该研究作了题为“不对称催化:令人鼓舞的手性”的报道。自Synfacts杂志创刊以来,课题组发表的论文有近1/2被作为Hightlight论文进行了高度评价和推荐。在对不对称氯胺化反应介绍中,Synfacts杂志评论道“The catalyst system achieves nearly perfect diastereoselectivity, and very high levels of enantioselectivity”该工作同时也被Synform杂志详细介绍。所发表的研究工作被编入30余部专著,100余篇论文被评述性论文正面评述,被邀请在国际、国内会议上做特邀和邀请报告30余次。
新型C2-对称双氮氧化合物的设计合成及催化不对称反应研究
氮氧化合物是一类强极性和强供电子性质的化合物基于双功能、C2对称性、分子识别等策略,我们首次设计合成了新型高效的手性氮氧配体和催化剂库,C=OC=N键的亲核加成反应、C=C键的加成反应、环加成反应及开环反应等多种类型不对称催化反应中有优秀的活性和立体选择性;其中利用手性氮氧-金属络合物催化剂首次实现了如Roskamp反应、α-重氮酯参与的不对称α-胺化反应,α,β-不饱和酮的卤胺化反应等形成碳杂原子键的不对称催化新反应
催化剂的设计思想:
早期的手性氮氧催化剂主要集中在喹啉类化合物衍生的双氮氧化合物,手性催化剂合成和修饰比较困难。N-杂芳环类氮氧衍生物的配位性质研究较多,但对叔胺类手性氮氧化合物的研究没有得到开展。我们从手性氨基酸出发,基于双功能和C2对称性策略合成了叔胺类手性双氮氧-酰胺类化合物,该类化合物含有Lewis碱和Brønsted酸中心的双功能催化特点;同时选择不同的桥链基团将氮氧-酰胺化合物连接可形成C2-对称的新型有机催化剂和手性配体,并具有原料廉价、合成简单、结构易调和使用方便等特点。突破了对配体刚性骨架的传统要求,是一类具有自主知识产权的新型优势手性配体
催化不对称反应研究:
(1)手性氮氧有机小分子催化剂催化不对称反应
首次将氮氧偶极化合物作为有机小分子催化剂,通过Lewis碱活化机理催化醛、酮和亚胺的腈化反应;利用氢键等非共价键作用催化羰基化合物的α-卤代反应,获得了高的催化活性和高的对映选择性,为含手性仲碳和季碳的二胺、氨基酸和α-卤代羰基化合物合成发展了简单实用的方法。
(2)手性氮氧-金属络合物催化剂催化不对称反应
氮氧偶极化合物具有强配位能力,可与20多种金属化合物如In(III)Sc(III)Yb(III)Y(III)Nd(III)La(III)Sm(III)Ni(II)Cu(I)Cu(II)Mg(II)Fe(II)Co(II)Ag(I)形成手性金属络合物催化剂,已在30余类不对称催化反应中获得优异结果,包括5类不对称催化有机新反应。反应和底物普适性强,催化效率高,一些反应中催化剂用量可降低至0.001mol%;许多反应可获得>99% ee的对映选择性;反应在温和条件下可扩大至克级规模,操作简单方便。

代表反应如下:

手性氮氧In(III)络合物催化剂
催化Danishefsky型二烯和Brassard二烯衍生物与醛或醛亚胺的不对称杂-DielsAlder反应、环氧化物开环反应、酮的烯丙基化反应。
催化不对称杂-Diels–Alder反应合成手性二氢吡喃酮
采用手性双氮氧酰胺配体与InIII组成的催化剂,实现了醛与Danishefsky型二烯的高效不对称催化杂Diels–Alder反应,可获得92–99% ee的对映选择性。制备好的催化剂存放六个月仍保持活性,广泛适用于多个底物,可放大到克级规模;以正戊醛为底物,高效高选择性合成了重要手性中间体triketide。研究工作被Nature新闻刊物Nature China做了题为“不对称催化:令人鼓舞的手性”的Research Highlights报道;英国皇家化学会杂志Chemistry World作了“Esters made easy with indium”的专题报道,不对称催化领域的著名有机化学家、芝加哥大学教授H. Yamamoto评价:“为金属铟化学开辟了新方向”。

手性氮氧Sc(III)络合物催化剂
首例醛与重氮酯的Roskamp反应、羰基α-位与重氮酯的直接胺化反应和扩环反应,首例α,β-不饱和化物的碘-胺化、溴-胺化和氯-胺化反应,3-取代氧化吲哚参与的Aldol反应及胺化反应,丙二酸酯与α,β-不饱和羰基化合物的Michael加成反应,吲哚与丙二酸亚烷基酯、查儿酮等的FriedelCrafts反应,三组分一锅法亚胺的不对称烯丙基化反应,KabachnikFields反应,Mannich型反应,氮杂-DielsAlder反应和反电子需求的氮杂-DielsAlder反应,α-酮酸酯与重氮酯的直接Aldol反应,环氧化反应及环氧开环反应,BV氧化促进取代环酮的去对称化和动力学拆分,酮的KBH4还原反应,串联反应等。
催化不对称Roskamp反应合成β-酮酸酯
催化不对称Roskamp反应需要解决化学选择性和立体选择性的控制、产物的消旋化问题,极具挑战性。手性氮氧–Sc(OTf)3络合物催化剂实现了首例高效高选择性的催化不对称Roskamp反应,催化剂用量可降低至0.05 mol%。研究工作发表后(J. Am. Chem. Soc. 2010, 132, 8532–8533),被编入专著Organic Synthesis Based on Name Reactions(第三版, 2011, Elsevier出版社, p 408),并冠以作者姓氏命名为“Roskamp–Feng反应”(Feng即冯小明)。
催化α-重氮酯与酮的不对称α-胺化反应
在取代α-重氮酯衍生物与酮的不对称催化反应研究中,实现了首例α-重氮酯作为亲电试剂参与的不对称催化反应,该研究丰富了重氮化学的研究内容,为酮的直接α-胺化反应提供了新方法。
催化α-重氮酯与滇红的不对称扩环反应
    在取代α-重氮酯衍生物与滇红及其衍生物的不对称催化反应研究中,采用手性氮氧–Sc(OTf)3络合物催化剂,在温和条件下实现了不对称扩环反应,主要得到1,2-芳基迁移产物喹啉酮衍生物。
催化不对称卤胺化反应
烯酮的双官能团化反应需要解决反应的区域选择性和反应中间体的消旋化等问题。在手性氮氧-Sc(OTf)3络合物催化剂条件下发现:烯酮、NBS和TsNH2的三组分反应能区域专一性发生溴胺化反应,产物α-溴-β-胺基酮的对映选择性高达99% ee。进一步将催化剂用于反应活性很低的氯胺化反应。通过对氯胺化反应活性试剂的分析,发现TsNH2和TsNCl2在4 ?分子筛存在条件下,能有效生成活性TsNHCl试剂,从而有效参与氯胺化反应。在0.05 mol%手性氮氧-Sc(OTf)3络合物催化剂条件下,能高收率和高立体选择性地得到α-氯-β-胺基酮类衍生物,该催化剂体系同样可被用于高效合成光学活性α-碘-β-胺基酮类衍生物。
催化不对称反电子需求杂-Diels-Alder反应
在手性氮氧-Sc(OTf)3络合物催化剂条件下,醛、邻羟基苯胺和环戊二烯能发生三组分“一锅法”的反电子需求氮杂-Diels–Alder反应得到手性环戊烯并四氢喹啉衍生物,对不同取代的芳基亚胺能获得62-99%的收率、90:10-98:2的顺反比和90->99% ee。该催化剂还能用于α-烷基苯乙烯为亲二烯体的不对称Povarov反应,合成含有季碳中心的四氢喹啉衍生物。
催化不对称烯酮的环氧化反应
手性氮氧-Sc(OTf)3络合物催化剂能有效催化查儿酮及其衍生物与双氧水(30%水溶液)的环氧化反应,获得高达99% ee的对映选择性。催化体系水容忍性高,反应条件温和,无需添加剂。
催化不对称环酮的Baeyer–Villiger氧化反应
利用手性氮氧-Sc(OTf)3络合物催化的不对称BV反应,实现了潜手性环己酮和环丁酮的去对称化,在乙酸乙酯为溶剂条件下能高收率和高对映选择性得到烷基和芳基取代的内酯衍生物。在2-芳基取代环己酮的动力学拆分反应中,加入一定量的Al(OiPr)3能促进BV氧化有效进行。有趣的是,在该催化剂条件下,(R)-构型的酮氧化得到非正常位置迁移的3-芳基-e-内酯,而仅有很少正常的BV氧化产物生成,且(S)-构型的酮能以很高的光学纯度回收。
催化KBH4对酮的不对称还原反应
    手性氮氧–Sc(OTf)3络合物催化α,β-不饱和酮和简单酮的不对称还原反应,仅需0.45摩尔当量的aq. KBH4还原剂,即能在温和条件,1.5小时内获得很好的收率和对映选择性。

手性氮氧Mg(II)络合物催化剂
催化不对称羰基杂烯反应
手性双氮氧–Mg(OTf)2能有效催化三氟甲基丙酮酸酯与烷基烯试剂的不对称羰基-烯反应以及滇红与α-甲基烯醚的不对称杂-烯反应,获得高达99% ee的对映选择性。其中4,6-二溴滇红衍生物与α-甲基烯醚的不对称杂-烯反应可高效合成药物分子(R)-convolutamydine A,收率高达86%,对映选择性97% ee。

手性氮氧La(III)络合物催化剂
催化硝基烯和硝基烷烃的Michael加成反应和硫杂-Michael加成反应。
催化不对称硫杂-Michael加成反应
在查儿酮类似物与巯基乙酸酯的杂-Michael加成反应中,我们发现:手性双氮氧–La(III)络合物能给出优异的结果,即使将催化剂用量降低至0.01 mol%,可在20毫摩尔规模得到中等收率和93% ee。催化剂表现出很强的手性放大效应,在2% ee光学纯度的氮氧配体,1 mol% 催化剂用量条件下即可催化反应得到高光学纯的手性含硫化合物。

手性氮氧Ni(II)络合物催化剂
催化活化α,β-不饱和羰基化合物的分子内氧杂-Michael加成反应、不对称羰基化物ene反应等。
催化不对称分子内氧杂-Michael加成反应
手性双氮氧–Ni(II)络合物能有效催化水杨醛衍生的活化α,β-不饱和羰基化合物的分子内氧杂-Michael加成反应,收率高达99%,对映选择性高达98% ee,为手性类黄酮类化合物的合成提供了一种有效方法。
催化不对称羰基ene反应及串联Prins反应
利用双氮氧–Ni(II)络合物催化乙醛酸酯和苯甲酰醛类化合物与α-甲基苯乙烯等的不对称烯反应,可高对映选择性得到高烯丙醇类化合物;以此为原料与醛进一步发生Prins反应,可高立体选择性地得到四氢吡喃衍生物;在FeCl3/TBSCl催化条件下化学专一性的生成4-羟基取代产物。

手性氮氧金属络合物催化反应中金属和溶剂调控的对映选择性反转现象
手性配体中非手性取代基的电子和位阻效应以及中心金属可调控反应的立体选择性,对催化剂结构和反应机理也进行了系统深入研究。
催化不对称Michael加成反应
在吡唑啉-5-酮衍生物与4-羰基-4-苯基丁烯酸酯的共轭加成反应中发现了中心金属和溶剂调控反应立体选择性的有趣现象。采用手性氮氧–Y(OTf)3络合物催化剂,二氯甲烷为溶剂条件下,主要得到(S,S)-构型产物;而手性氮氧–Sc(OTf)3络合物催化剂在乙醇溶剂中,主要得到(R,R)构型的产物。

新型手性双功能有机小分子催化剂的设计合成及催化不对称反应研究
基于双功能催化剂的设计策略,利用廉价易得的的手性氨基酸,我们设计合成了手性氨基酸盐催化剂、基于双哌啶骨架的二胺类催化剂和手性胍-酰胺类催化剂。
含双哌啶骨架的伯胺-仲胺催化剂能高效催化活化酮如a-羰基磷酸酯、a-羰基羧酸酯,a,a-二烷氧基酮的不对称aldol反应,获得高达97%收率和98% ee。还能有效催化各种酮如环己酮、丙酮和各种芳香酮与硝基烯衍生物和烯酯的Michael加成反应。一些反应能在无溶剂条件或水相中进行。
胍是最强的Lewis碱,能作为有机碱催化剂;也能与质子酸形成胍盐催化剂,通过分子间氢键作用活化底物或试剂。我们将胍和酰胺基团结合起来,发展了一类新的手性胍-酰胺催化剂的合成方法。将胍-酰胺化合物作为有机小分子催化剂用于b-酮酯对硝基烯的1,4-加成产物中可获得高达99%的收率,>99:1 dr 和97% ee,为手性雷米普利类似物的合成提供了有效方法。在吖内酯与查儿酮的加成反应中发现:在C2-对称的双胍-酰胺催化剂条件下,主要得到反电子需求杂-Diels-Alder反应产物——含季碳中心的δ-内酯衍生物,仅有少量的常规Michael加成产物,立体选择性高达99%ee,这也是首例查儿酮参与的直接不对称杂-Diels-Alder反应。采用水杨醛衍生的亚胺与吖内酯发生Mannich-酰胺化串联反应能一步合成3,4-二氨基苯并二氢吡喃-2-酮衍生物反应中发现,当采用单胍-酰胺催化剂时,产物主要以顺式为主,对映选择性可达9096% ee;当采用C2对称胍-酰胺形成的BArF4盐为催化剂时,产物主要以反式为主,对映选择性可达8899% ee该催化剂在不对称Mannich型反应中也有很好的应用。
氨基酸与碱金属形成的盐可高效催化酮的不对称硅腈化反应以及酮与硝基烯的Michael加成反应。该类催化剂廉价易得,可回收循环使用。
双功能、双活化、组合催化策略和其它金属络合物催化不对称反应研究
将廉价易得的的手性配体、非手性配体和金属化合物有机组合起来,构建具有优秀手性环境的双功能催化剂。发现组合催化剂生物碱-Ti(OiPr)4-取代联苯酚在C=O键、C=N键甚至是相对惰性的C=C键的氰基加成反应中取得了优异的结果,也是该类反应中唯一能催化C=O键、C=N键甚至是相对惰性的C=C键的不对称腈化反应最具广谱性和高选择性的催化剂体系。对催化剂结构和反应机理研究发现,在催化剂的形成中伴随着不对称活化作用。
将双活化策略用于酮的不对称硅腈化反应,催化剂用量可低至万分之五,仍能获得优异的结果,是目前最好结果的最低催化剂用量记录保持者,美国Amgen医药公司也成功地将该方法用于治疗糖尿病药物11β-羟甾类脱氢酶抑制剂AMG221的合成中。
